研究内容及意义
自然条件下大多病毒借助虫媒传播,不但涉及病毒本身,而且涉及宿主、媒介以及宿主和媒介体内的共生微生物,是一个多因素的复杂生物学过程。目前,世界范围内对虫媒这一类自然宿主生物传播病毒的生物学基础和分子生物学机制的认识依然不足。多数虫媒病毒病至今缺乏像临床药物和疫苗等在内的有效控制手段,在人及动植物领域,对于新发突发虫媒病毒病均只能在暴发后采取被动控制措施。因此,通过多学科交叉合作,系统深入了解虫媒传播病毒生命循环的基本过程和机制迫在眉睫,对虫媒病毒传播的生物学基础和病毒适应跨界宿主机制进行解析,将防控虫媒病毒病的关口前移,研发抗病毒手段势在必行。本研究组在国家自然基金委重大、重点、科技部重点研发及中科院先导等项目大力支持下主要开展以下三个研究方向的工作:
1. 虫媒病毒自然宿主的免疫适应。我们致力于将微观分子生物学、遗传学、媒介昆虫神经生物学技术,发掘、解析和利用抗虫媒病毒相关的代谢重塑机制,发展新型广谱抗病毒化合物。本课题组最近发现入侵双生病毒具有的全新生态学功能,双生病毒感染棉花等植物不仅促进媒介昆虫烟粉虱,而且阻止非介体竞争对手棉铃虫和蚜虫的生态功能,促进双生病毒入侵危害。具体研究成果参见近期代表工作1。
2. 植物抵御虫媒病毒免疫网络解析。最经济有效的防控虫媒植物病毒病的方法是获得抗性品种,解析宿主的抗性机制至关重要。通常实验研究在实验室条件下,植物对一种抗性(病毒或者昆虫)机制,这种研究往往不符合自然条件的病毒传播规律,因为80%的植物病毒在自然条件下由媒介昆虫传播,而作物往往受到虫媒外加病毒的双重复杂胁迫。我们研究植物应对复杂生物胁迫的分子、细胞机制,并通过先进生物技术手段发展抗虫媒病毒传播和传播的新方法、新种质材料,有效防控新发病毒疾病提供了理论基础和绿色应用生物技术,获省部级科技进步奖一项。(参见近期代表工作2)
3. 虫媒柑橘黄龙病的宿主抗病机制及抗病生物技术。目前已知有超过300种细菌病害经由昆虫媒介传播,包括引起黄龙病的柑橘韧皮部杆菌(liberibacter)、螺原体、植原体、苛养木杆菌等,由于多数作物中缺乏抗性基因,主要通过杀灭媒介昆虫来防控。我们通过利用人工智能(AI)结合时空组学等多种前沿方法,高通量从野生作物种质资源中挖掘新型抗病(感病)基因,解析抗病(感病)机制,发展新型抗病生物技术。目前已申请或者获批多项中国和国际发明专利。

近期代表工作1:双生病毒操纵植物免疫来抑制非介体昆虫。棉花等植物被双生病感染后,大幅度提宿主植物抵御农业重大害虫—棉铃虫和蚜虫的能力,抗棉铃虫的能力甚至比转杀虫Bt毒基因的抗虫棉还好。进一步研究发现双生病毒利用了高等双子叶植物中保守的免疫抗性基因WRKY20调控的植物的免疫平衡制约机制,显著减少植物维管束组织(双生病毒侵染部位和烟粉虱取食部位)中抗虫化合物的积累,如芥子油苷等,促进传毒介体烟粉虱的种群增长。但是会提高非维管束组织如叶肉细胞中脂肪族芥子油苷水平,并激活植物体内阿司匹林类药物-水杨酸的产生,从而抑制棉铃虫和蚜虫—烟粉虱的主要竞争对手的生长,进一步促进了与烟粉虱的互惠共生,加速了病毒在全球范围内的入侵,造成了严重的粮食安全和生物安全问题。(Science Advances, 2019)
近期代表工作2:光调控病毒-昆虫-植物三者互作的分子机制。利用单色光LED灯箱探究不同光调控植物的抗虫和抗病途径,发现红光通过PIFs蛋白调控萜烯类化合物的合成起到抗虫的作用(图A, PLoS Pathogens, 2021)。红外光通过促进PIF4蛋白积累激活RDR6/AGO1的表达,通过影响RNAi途径调控植物抗病反应(图B, Plant Com, 2023),以上研究为利用单色LED灯绿色防控虫媒病毒病害提供理论基础。
近期代表工作3:本课题组结合基因组、泛基因组和性状功能分析挖掘甘蔗高产高糖抗病关键调控基因,利用新型基因组编辑技术,突破甘蔗高效基因编辑瓶颈,进行多点多年田间育种评价、大数据辅助品种选育等现代生物技术,快速创制高产量高糖分甘蔗优良新种质,全面提升甘蔗的产量、含糖量和抗逆性,突破甘蔗产业链发展瓶颈的核心种质创建问题,建立高复杂度基因组经济作物育种新范式。
